台灣行政院長蘇貞昌年前請辭，總統府1月25日晚間證實，確定由前副總統陳建仁接棒，總統蔡英文27日召開記者會正式宣布。
去年11月，執政的民進黨在地方選舉大敗後，人民要求行政院長蘇貞昌下台的呼聲很高。蘇貞昌最後在立法院通過中央政府總預算後，1月19日向蔡英文總統請辭。
中央社報導，總統府發言人張惇涵25日晚間表示，針對新任閣揆人選，蔡英文當天下午邀請陳建仁就重大國政議題交換意見，並正式徵詢他後定案，隨後立即告知蘇貞昌及副總統賴清德。
張惇涵指出，蔡英文預計27日上午10時於總統府召開記者會正式宣布；執政團隊的目標是穩定政局、開創新局，並在過年後，讓新的內閣團隊迅速就位，讓各項國政無縫接軌、順利推動。
陳建仁現年71歲，篤信天主教，是國際知名流行病學專家。他數月前加入民進黨。他在擔任副總統之前，曾任行政院衛生署署長、行政院國家科學委員會主任委員、中研院副院長等職務，副總統任內曾兼任國家年金改革委員會與總統府人權諮詢委員會召集人。
陳建仁的學術成就斐然，是國際知名砷中毒專家，相關研究之論文系列被認定是環境流行病學經典著作；另一著名貢獻是慢性B型肝炎，曾率領團隊進行全球規模最大、時間最長的肝炎病毒追蹤研究，發現預測末期肝病重要分子標幟。
此前，民進黨立委許智傑回應媒體詢問時表示，若陳建仁接任行政院長，他致上最高祝福，並期許新內閣是全民內閣，帶領人民告別疫情，發展經濟照顧弱勢。
他指出，陳建仁有三大特質，包括學者出身，社會形象佳；過去擔任許多改革的召集人，溝通力強；外交、宗教界人脈廣，具有宗教家照顧弱勢的情懷。
國民黨立委江啟臣則於媒體群組發表看法稱，人民想問的是新內閣打算做什麼，哪一項可以腳踏實地做好；期許陳建仁能以人民利益為念，做實事，多包容，少政治，相信天主應該也是這樣期許。
台灣民眾黨立委張其祿發布新聞稿指出，目前民眾真正在乎的是「問題解決」，未解的國計民生問題仍然繁多，皆須有高效度的行政院長及各部會首長儘速解決問題。
張其祿表示，若由陳建仁出任行政院長，期望能夠以快速且務實的行動力，加緊處理國家當前面對的經濟、國防、兩岸問題及民生挑戰。
